چکیده
The most widely used method for determination of sexual compatibility between two isolates of Cryphonectria parasitica (C.p.) involves co-inoculation of the strains into dormant American chestnut stems. The method is timeconsuming (requiring an average 8-10 weeks for observable perithecial necks of the fungus in chestnut bark) and needs a ready supply of suitably-sized dormant stem pieces. The objective of this project was to find an artificial laboratory medium that would facilitate sexual reproduction in C. parasitica in a timely manner.

Low diversity of vegetative compatibility types and mating type of Cryphonectria parasitica in the southern Balkans
The diversity of vegetative compatibility (vc) types and mating type was estimated in populations of the chestnut blight fungus, Cryphonectria parasitica, throughout Macedonia and from selected areas in Greece. Nearly all of the 786 isolates (94%) from Macedonia were in a single vc type, EU-12; all 379 isolates from Greece were EU-12. Only six of 20 populations in Macedonia had more than one vc...
متن کاملAphelenchoides hylurgi as a carrier of white, hypovirulent Cryphonectria parasitica and its possible role in Hypovirulence spread on blight-controlled american chestnut trees.
Individual nematodes were isolated from American chestnut blight-controlled cankers to determine if they were carriers of biocontrol (hypovirulent) isolates of the chestnut blight fungus, Cryphonectria parasitica. These hypovirulent isolates have a white fungal colony phenotype due to infection by the virus CHV1. Of 1,620 individual Aphelenchoides hylurgi isolated, 29.4% carried propagules of t...
متن کاملInvasion history and demographic pattern of Cryphonectria hypovirus 1 across European populations of the chestnut blight fungus
We reconstructed the invasion history of the fungal virus Cryphonectria hypovirus 1 (CHV-1) in Europe, which infects the chestnut blight fungus Cryphonectria parasitica. The pattern of virus evolution was inferred based on nucleotide sequence variation from isolates sampled across a wide area in Europe at different points in time. Phylogeny and time estimates suggested that CHV-1 was introduced...
متن کاملPotential of Local Hypovirulent Strains of Cryphonectria parasitica for Biological Control of Chestnut Blight
Cryphonectria parasitica, introduced in the NE of Portugal since 1989, is now an important and widespread pathogen of chestnut (Castanea sativa Mill.) which causes losses in chestnut fruit production and led to decline and death of many chestnut trees. Hypovirulence, a virus mediated attenuation of fungal pathogenesis, has not been frequent until now in Portugal but recently some cases of heali...
متن کاملMultilocus PCR Assays Elucidate Vegetative Incompatibility Gene Profiles of Cryphonectria parasitica in the United States.
Chestnut blight is a devastating disease of Castanea spp. Mycoviruses that reduce virulence (hypovirulence) of the causative agent, Cryphonectria parasitica, can be used to manage chestnut blight. However, vegetative incompatibility (vic) barriers that restrict anastomosis-mediated virus transmission hamper hypovirulence efficacy. In order to effectively determine the vegetative incompatibility...
